Eyewitnesses tell Sky News the main hospital in Raqqa has been closed to the public - suggesting the death of a senior IS leader.
An airstrike targeting the British IS militant known as "Jihadi John" has been carried out in Syria - and the US is "99% sure we got him".
Activists in the IS stronghold said they counted 14 airstrikes between 11.51pm and midnight on Thursday.
They reported that one of the first missiles targeted a car near the city's Islamic court - but the area was closed off by militants to prevent anyone from approaching.
Eyewitnesses in Raqqa told Sky News Jihadi John - real name Mohammed Emwazi - was taken to hospital after the strike, with IS claiming he was injured but still alive.
Although the official line from IS leaders is that Emwazi survived, locals told Sky the hospital has since been closed to the public and claimed this only happens when a senior militant has been killed.
Pentagon press secretary Peter Cook confirmed the strike was directed at Emwazi but said it was not clear whether he had been killed.
We are assessing the results of tonight's operation and will provide additional information as and where appropriate," he said.
However, a senior US defence official told Fox News: "We are 99% sure we got him. We were on him for some time."
Another source told ABC News Emwazi was blown up in a "flawless" and "clean hit".....etc.

During the summit, “I provided examples based on our data on the financing of different Islamic State (IS, formerly ISIS/ISIL) units by private individuals. This money, as we have established, comes from 40 countries and, there are some of the G20 members among them,” Putin told the journalists.
Putin also spoke of the urgent need to curb the illegal oil trade by IS.
"I’ve shown our colleagues photos taken from space and from aircraft which clearly demonstrate the scale of the illegal trade in oil and petroleum products," he said.
“The motorcade of refueling vehicles stretched for dozens of kilometers, so that from a height of 4,000 to 5,000 meters they stretch beyond the horizon," Putin added, comparing the convoy to gas and oil pipeline systems.
It’s not the right time to try and figure out which country is more and which is less effective in the battle with Islamic State, as now a united international effort is needed against the terrorist group, Putin said.
Putin reiterated Russia’s readiness to support armed opposition in Syria in its efforts to fight Islamic State.
"Some armed opposition groups consider it possible to begin active operations against IS with Russia's support. And we are ready to provide such support from the air. If it happens it could become a good basis for the subsequent work on a political settlement,” he said.
“We really need support from the US, European nations, Saudi Arabia, Turkey, Iran,” the president added.
Putin pointed out the change in Washington’s stance on cooperation with Moscow in the fight against the terrorists.
"We need to organize work specifically concentrated on the prevention of terrorist attacks and tackling terrorism on a global scale. We offered to cooperate [with the US] in anti-IS efforts. Unfortunately, our American partners refused. They just sent a written note and it says: ‘we reject your offer’,” Putin said.
“But life is always evolving and at a very fast pace, often teaching us lessons. And I think that now the realization that an effective fight [against terror] can only be staged together is coming to everybody,” the Russian leader said.
Suddenly, they're all desperate to be seen with Putin, after all the hot air and bombast at the previous G20.

The Obama White House is said to have reached out to Putin staffers wanting to have a quick sit-down moment just prior to the beginning of the G20 Summit hosted this week by Muslim Brotherhood advocate and Turkish Dictator President, Recep Tayyip Erdogan. It was a blatant attempt at a photo-op said to have been suggested and strongly pushed for by longtime Senior White House Adviser, Valerie Jarrett.
Regardless of personal views world leaders might have of his tactics, Russia’s Putin is now seen as the single most influential player in the war on Islamic terror and the Obama White House was desperate to try make itself appear equally worthy of the fear and respect the world has for Putin’s determination to wipe ISIS off the face of the planet.
The result was this photo being aggressively pushed for widespread publication by the Obama White House.
What the White House won’t be pushing is the apparent reaction Putin had to Barack Obama’s self-serving overtures. He was seen laughing among his staff shortly after the already famous “coffee table” moment, and looking over at Mr. Obama with what is being described as “dismissive derision.”

Meanwhile, the Russian military launched airstrikes against the ISIL stronghold in Raqqa with cruise missiles from the Mediterranean Sea.
On Tuesday, the France’s Le Monde newspaper, citing a French official, said that the Russian military has launched airstrikes against the ISIL stronghold of Raqqa in Syria with cruise missiles from the Mediterranean Sea.
The information that Russia struck the ISIL positions in Raqqa was also reported by a senior French government source.
US officials also confirmed on Tuesday that Moscow conducted a significant number of strikes in Syria using both sea-launched cruise missiles and long-range bombers.
In the meantime, different sources said that the French fighter jets carried out many sorties over the ISIL positions in recent day and targeted them heavily with scores of bombs and missiles in reprisals for the Paris terror attacks by ISIL.
Also on Tuesday, French President Francois Hollande told the French parliament’s emergency meeting in Versailles that the French nuclear-powered Charles de Gaulle aircraft carrier will leave the port of registration on November 19 to join the operation in Syria.

On Tuesday, the France’s Le Monde newspaper, citing a French official, said that the Russian military has launched airstrikes against the ISIL stronghold of Raqqa in Syria with cruise missiles from the Mediterranean Sea.
The information that Russia struck the ISIL positions in Raqqa was also reported by a senior French government source.
US officials also confirmed on Tuesday that Moscow conducted a significant number of strikes in Syria using both sea-launched cruise missiles and long-range bombers.
In the meantime, different sources said that the French fighter jets carried out many sorties over the ISIL positions in recent day and targeted them heavily with scores of bombs and missiles in reprisals for the Paris terror attacks by ISIL.
Also on Tuesday, French President Francois Hollande told the French parliament’s emergency meeting in Versailles that the French nuclear-powered Charles de Gaulle aircraft carrier will leave the port of registration on November 19 to join the operation in Syria.

I'm watching Fox just now; they seem awfully frustrated that all this mayhem and shooting is taking place in Paris, and is nothing to do with Washington.
And that Russia, France and others are pulverising ISIS, and it's nothing to do with Washington.
I suspect that those involved in pulverising ISIS don't actually trust the Obama regime - and why should they? No-one in their right mind does.
I'm watching Fox just now; they seem awfully frustrated that all this mayhem and shooting is taking place in Paris, and is nothing to do with Washington.
